    Shuttle active thermal control system development testing. Volume 6: Water ejector plume tests

    Results are given of vacuum testing of nozzles designed to eject water vapor away from the space shuttle to prevent contamination of the spacecraft surfaces and payload. The water vapor is generated by an active cooling system which evaporates excess fuel cell water to supplement a modular radiator system (MRS). The complete heat rejection system including the MRS, flash evaporator or sublimator and nozzle were first tested to demonstrate the system operational characteristics. The plume tests were performed in two phases and the objectives of this test series were: (1) to determine the effectiveness of a supersonic nozzle and a plugged nozzle in minimizing impingement upon the spacecraft of water vapor exhausted by an active device (flash evaporator or sublimator); and (2) to obtain basic data on the flow fields of exhaust plumes generated by these active devices, both with and without nozzles installed

    SRC-FAMILY TYROSINE KINASES PARTICIPATE IN THE REGULATION OF MAMMALIAN OOCYTE MATURATION AND ZYGOTIC DEVELOPMENT

    Multiple signaling pathways act during ovulation to ensure that the oocyte is prepared for embryonic development. We combined pharmacological and cell biological methods to study the role of Src-family kinases (SFKs) during oocyte maturation, fertilization and embryonic cleavage in mice. Active SFKs localize to spindle microtubules and the cell cortex. Fyn kinase is the primary SFK in mouse oocytes. Disruption of Fyn kinase causes abnormalities in chromatin separation and spindle formation. Site specific tyrosine phosphorylation events in the egg cortex near chromatin are altered by the conditions under which oocyte in vitro maturation occurs. Thus, previously unanticipated functions for SFKs are identified that mediate spatial and temporal remodeling of cytoskeleton and cell cycle during oocyte maturation and early development. These findings bear directly on human assisted reproductive technology with the discovery of the role of this pathway in maturation and embryonic cleavage of mammalian oocytes

    Accuracy of the QUAD4 thick shell element

    The accuracy of the relatively new QUAD4 thick shell element is assessed via comparison with a theoretical solution for thick homogeneous and honeycomb flat simply supported plates under the action of a uniform pressure load. The theoretical thick plate solution is based on the theory developed by Reissner and includes the effects of transverse shear flexibility which are not included in the thin plate solutions based on Kirchoff plate theory. In addition, the QUAD4 is assessed using a set of finite element test problems developed by the MacNeal-Schwendler Corp. (MSC). Comparison of the COSMIC QUAD4 element as well as those from MSC and Universal Analytics, Inc. (UAI) for these test problems is presented. The current COSMIC QUAD4 element is shown to have excellent comparison with both the theoretical solutions and also those from the two commercial versions of NASTRAN that it was compared to

    Mammalian oocytes are targets for prostaglandin E2 (PGE2) action

    <p>Abstract</p> <p>Background</p> <p>The ovulatory gonadotropin surge increases synthesis of prostaglandin E2 (PGE2) by the periovulatory follicle. PGE2 actions on granulosa cells are essential for successful ovulation. The aim of the present study is to determine if PGE2 also acts directly at the oocyte to regulate periovulatory events.</p> <p>Methods</p> <p>Oocytes were obtained from monkeys and mice after ovarian follicular stimulation and assessed for PGE2 receptor mRNA and proteins. Oocytes were cultured with vehicle or PGE2 and assessed for cAMP generation, resumption of meiosis, and in vitro fertilization.</p> <p>Results</p> <p>Germinal vesicle intact (GV) oocytes from both monkeys and mice expressed mRNA for the PGE2 receptors EP2, EP3, and EP4. EP2 and EP4 proteins were detected by confocal microscopy in oocytes of both species. Monkey and mouse oocytes responded to PGE2 as well as agonists selective for EP2 and EP4 receptors with elevated cAMP, consistent with previous identification of EP2 and EP4 as Gαs/adenylyl cyclase coupled receptors. Incubation of mouse GV stage oocytes with PGE2 delayed oocyte nuclear maturation in vitro, but PGE2 treatment did not alter the percentage of mouse oocytes that fertilized successfully. PGE2 treatment also decreased the percentage of monkey oocytes that resumed meiosis in vitro. In contrast with mouse oocytes, the percentage of monkey oocytes which fertilized in vitro was lower after treatment with PGE2. Monkey oocytes with intact cumulus showed delayed nuclear maturation, but fertilization rate was not affected by PGE2 treatment.</p> <p>Conclusions</p> <p>Monkey and mouse oocytes express functional PGE2 receptors. PGE2 acts directly at mammalian oocytes to delay nuclear maturation. Surrounding cumulus cells modulate the effect of PGE2 to alter subsequent fertilization.</p

    Paradox reconsidered: Methane oversaturation in well-oxygenated lake waters

    The widely reported paradox of methane oversaturation in oxygenated water challenges the prevailing paradigm that microbial methanogenesis only occurs under anoxic conditions. Using a combination of field sampling, incubation experiments, and modeling, we show that the recurring mid-water methane peak in Lake Stechlin, northeast Germany, was not dependent on methane input from the littoral zone or bottom sediment or on the presence of known micro-anoxic zones. The methane peak repeatedly overlapped with oxygen oversaturation in the seasonal thermocline. Incubation experiments and isotope analysis indicated active methane production, which was likely linked to photosynthesis and/or nitrogen fixation within the oxygenated water, whereas lessening of methane oxidation by light allowed accumulation of methane in the oxygen-rich upper layer. Estimated methane efflux from the surface water was up to 5 mmol m(-2) d(-1). Mid-water methane oversaturation was also observed in nine other lakes that collectively showed a strongly negative gradient of methane concentration within 0-20% dissolved oxygen (DO) in the bottom water, and a positive gradient within \u3e= 20% DO in the upper water column. Further investigation into the responsible organisms and biochemical pathways will help improve our understanding of the global methane cycle

    Psychometric Characteristics of the Connor-Davidson Resilience Scale (CD-RISC) in Postpartum Mothers with Histories of Childhood Maltreatment

    Background: There is increased awareness that resilience serves as a protective factor against adverse psychophysiological sequelae in the context of stress. However, there are few instruments to assess this construct in adult populations. The Connor-Davidson resilience scale (CD-RISC) has been developed to assess adaptation following stress exposure. While this instrument has previously demonstrated impressive reliability and construct validity, prior research has not supported the consistency of the originally described factor structure. There is also limited evidence regarding the measurement of resilience in the context of cumulative stress exposure. Objectives: This research explores the psychometric properties of the CD-RISC in mothers with childhood histories of maltreatment Materials and Methods: Postpartum women who endorsed a history of childhood abuse or neglect (N = 141) completed the CD-RISC, the childhood trauma questionnaire and other surveys measuring positive and negative health and functioning. We calculated descriptive statistics with percentage counts and means as appropriate. Internal reliability was evaluated by Cronbach’s alpha and the calculation of item-to-total score correlations. Parallel analysis (PA) was utilized to derive the number of retained factors. Results: A recent parenting transition concomitant with a history of maltreatment was associated with lower CD-RISC scores. Internal reliability and concurrent validity analyses were satisfactory and consistent with predicted hypotheses. Exploratory factor analysis (EFA) supported a four-factor model of resilience with this population. Conclusions: This research offers further evidence of the reliability and validity of the CD-RISC. Further, the results of the EFA with parallel analysis offer an empirically-driven derivation of factors for this population

    Engaging Children in Design Thinking Through Transmedia Narrative (RTP)

    This paper presents the implementation of Imaginative Education (IE) pedagogy for creating a transmedia learning environment that engages children in learning about engineering design. IE uses narrative to engage learners\u27 imaginations; helps them master the cognitive tools necessary for progressing to higher levels of understanding; and helps them structure what they learn in meaningful ways. Included in the paper is an introduction to IE pedagogy and the use of transmedia in education; an overview of the online learning environment called Through My Window (TMW) that we have developed for middle school children; and a detailed look at a learning adventure on engineering design called Trapped in Time. Assessment data collected by external evaluators shows that TMW positively impacted student interest in engineering and increased STEM identity. Preliminary results for the Trapped in Time learning adventure indicate improved understanding of engineering design
